I think most readers use one book per prompt, but it's completely up to you, there are no rules and no one will check up on what you read.
There is a post where people are sharing spreadsheets they created:

We do not maintain a "group spreadsheet" here, you make a copy for yourself and it's your own personal list.
There is also a post where members have shared the books they plan, and you can go back and edit your comment each time you complete one of the categories:

If there is a particular book you want to read but you can't figure out which category you could check off with it, you can post your question here:
/topic/show/...

At least, in theory I do. In practice I never actually get that far. But as has been said already, there are no rules. Do what works for you.

I used a mix of books I plan to reread and new books. I, also, have a different book for each prompt because I'm using this challenge to meet my goodreads book goal of 110 books read this year.

There is a listopia for each prompt that is linked in the first post of each prompt's discussion thread. If the books you are interested in are labeled as "Want to Read" in Å·±¦ÓéÀÖ, it is pretty easy to see the differently colored box or boxes as you scroll through the lists.

You can search the discussion posts for the title of the book and see if it comes up in any of the prompt threads. This is in the top right on a desktop browser. I don't know if you can search groups in the app.

You could also post in the monthly discussion and ask everyone there where the specific title you're thinking of reading might fit in the challenge. You're likely to get a few responses quickly there.
I posted about a book I have out from the library that's not for the challenge and got a bunch of prompt suggestions right away :-)

If you go the discussion thread, there's a "more" at the bottom of the list. Click that and you'll find the one for #2 (whichever that one is).


Apparently I'm a compulsive "completist" and once I have my list of books for the Challenge I just have to keep reading (also I read very fast and I'm retired). So, this is to announce that I have finished the Challenge (regular and advanced). I know in the past there has been a list of questions to answer when finished, but I don't see it yet so I'll try to answer the ones I can remember anyway.
My list number is 398. (My Å·±¦ÓéÀÖ shelf name is 2020 Popsugar under Judy Duer if you want to see reviews.)
My favorite prompt was "a book on a subject you know nothing about" The Ghost Map by Steven Johnson (about the Victorian era cholera plague in London)
My least favorite prompt was Read a Banned Book During Banned Books Week. I read a banned book, but didn't want to wait until September to complete the challenge, so I have pledged to read another banned book then to make up for it.
Will I do the challenge in 2021? I imagine so, barring some catastrophe. I would probably still have read this many books at this point in the year, but I like being encouraged to read things I might not have read otherwise.

Congratulations Judy! I'm impressed! Yeah, we usually set up a thread for completion of the entire challenge about midway through the year. Maybe we need to consider opening it earlier for our power readers ;)
